I am sitting waiting for a trailer to come pick me up as I type this... My bike started missing, one cylinder firing one not intermittently. Didn't matter how much load was on it low rpms or high rpms. It got real bad so I pulled into a gas station.. When I went to start it back up it only fired on one cylinder.. Ignition module? The coil has been replaced because it did something similar before and the coil tested bad so it was replaced.. Bike has ran fine until now..
Go to spark plugs first , then check for voltage leakage at plug wires in a dark room to check for leaking spark.
Check all signal wire connections to coils for looseness etc.
Use a cheap spark plug testor to check for proper spark at the offending cylinder . Come back with results.

No, not the speedo. And, ahhh... if your indy is suggesting the speedo is causing the rear cylinder to miss, ah... it's time to find a new indy.
When the bike is missing, that's when you need to diagnose it. Did you ever check the compression? Did you ever check for spark? Did you ever replace the rear spark plug? Did you swap the plug wires at the coil?
