Default carb getting no fuel

I have a 2000 lowrider with a mikuni carb on it. I took the petcock fuel filter out to replace it, followed clymer manual instructions, and replaced the filter. Put the fuel valve back on, went to start up and it just tried to start but sounded like it had no fuel to fire up. I drained the tank, took the pet **** back off and turned it from OFF to ON and RESERVE and the fuel came out of it at a normal rate. But when I hook the fuel line back up same problem. I have bypassed teh vacuum line and tried it that way and the stock way, still trying to start but not getting gas to the carb. Any ideas? Thanks for all help in advance

Try new fuel line from petcock to carb.

Tried that yesterday, still same problem. Any other ideas?

With air breather off, twist throttle and see if fuel is squirting at throat of carb. Recheck petcock to ensure no o-rings are displaced or pinched, stopping any fuel flow. Vacuum should not be an issue.

Sounds like maybe some dirt plugged the carb inlet. Pull the carb, remove the float and blow air backwards through the inlet. I would put a piece of hose on it in a bottle so you can see what comes out

Got it sloved! The problem was the carb inlet thanks for the help. I was at a loss to what the fix would be. Not very mechanically inclined, but trying to get better!
