Definitely Rt. 66 then head on East from Oklahoma or Missouri to Tennessee (Tail of the Dragon), North Carolina (Blue Ridge Parkway), Virginia (Skyline Drive) and ride on the back roads from Front Royal, VA through PA Mountains and upward to Northeast.

Another suggestion is you'd probably want to see Lake Placid (Site of 1980 Olympics), cross Lake Chaplain by Ferry to look for Loch Ness Monster, check out VT 108, Vermont (No center stripe) up in the mountains and Mount Washington (The World's Worst Weather) in New Hampshire before heading for your final destination.
