Ok, I've had my 2010 Ultra for a month now and have put a little over 1k on it. This is my sixth harley and third ultra. My observations so far are:
The brakes take some getting used too, they really grab quick.
The seat height has increased about 2" over the previous frame design.
The new frame feels much more rigid which I'm still getting used to.
There are no issues with the infamous "5th Gear noise" but the engine sounds like a sewing machine though.
I installed rinehart slip-ons on the first day and have to say that I'm disappointed. They only sound good when you're getting on it otherwise they sound about like stock at idle. They are also are starting to turn a bronze color from the heat.
There is significantly less heat near the right inner thigh whrere the cross-over used to be. But there seams to alot more heat now near my right ankle, I assume that is where the catalyst is located.
Other than that, I cannot tell the difference between my 2010 and the ones I had in previous years. I know that there have been several hundred improvements since 2006.

I loved my 06 and 07 ultras but am feeling the same about my 10. Like I said, it may just be a matter of getting used to riding a bagger again. My last bike was an 07 springer classic which I absolutely enjoyed but missed the functionality I had with my baggers; cruise, storage, tunes and most importantly comfort for mama. My biggest issue with my 10 is the height; even though I installed profile shocks, I'm still on my tip toes. I've experimented with 10 - 50 psi but feel like I'm bottoming out or a clunk whenever I hit a significant bump in the road. I put it on a lift and checked the fender well but and not see any evidence of the rear tire rubbing the fender so I'm led to believe there may be a design flaw with the rear suspension.
I forgot to add that the bike will not start while in gear. I parade often and have had me previous bikes cut off right in the middle of a parade, this could be a real pain to have your engine cut out and stop, find neutral, start and then go again...

My Limited starts is gear.
I do it all the time because it is hard to find netrual.
I have got 1300 miles on my 2010 Ultra and I think it's just a better bike than the 2007 Ultra I traded. The 2007 had 26000 miles at trade with no problems other than a starter replace under warranty. It had SE 255 cams, SERT & Rhinehart slipons.
The 2010 is much cooler other than the heat on right ankle area, it handles much better and just seems smoother on the road. I am following thread here about drilling cats out of pipes. Just had the SE air filter and download yesterday and that helped also.
My bike will start in gear though. Much improved, my 2 cents...
That's interesting; I've ran a stage one on every harley I owned except my 10 because the service manager says there are no down loads for 2010's. I would love to replace the football with a stage one.

Only if the kick stand is down and the clutch lever is not pulled in.
My first and only Harley is a '10 Limited, and I have no complaints whatsoever. The only thing that bugs me a little is I've had to pump up the rear shocks 3 times so far.

Definitely have to agree about the brakes.
Traded in an '07 GL1800 for my '10 Limited 3 weeks ago and on the ride home
the wife kept sliding into me at stops - those brakes are sensitive!
Had never been on a Harley before the Ultra I did a test ride on.
Took it to our GWRRA chapter meeting this month and I think it made lots of folks jealous :-)
