Just about anywhere on the parkway is a good ride. Two key things to keep in mind. I think the BRP is one of the most poorly marked roads I have ever been on. Also keep in mind that the speed limit is 35-45 and you will get a lot of slow drivers. This will impact your travel time.

There are a lot of slow driversbut there are a lot of pull offs and I find people will get off rather than have a Mc on their tail. Also when you consider there are no stop signs or lights that ups your 45mph average.

If it were me, I'd break up that Boone-Cherokee leg with an overnight stop in Asheville to allow more time to enjoy the rides on and around the BRP......
I live just outside Asheville (Leicester) and I can second this advice. It's solid. Tons of places to stay/eat/drink in Asheville as well.

The only thing I would add to this is if you are coming now (actually any time from August until the end of October) is to reserve your hotel room in advance in Asheville. They book up quickly in the fall.

...and there are a lot of people on the parkway when the leaves start to turn, so be extra careful.

Have a great trip! I'll probably pass a few of you going the other way!
