I picked up a computer program called Microsoft Streets and Trips 2006 (its less than $40). It is good for US & Canada. I am using it to plan a 2 week Canadian excursion in July/August. I have found it very easy to use and would recommend this.

From LI, I'd take NJ 23 to get to Port Jervis, then NY 97 N to Hancock. NY17 W to Corning to NY417 W to Salamanca. Pick up NY17 again W to just west of Jamestown. When you cross over Chautauqua Lake, take NY 394 N to US 5 E or US 20 E - US 5 will take you into Buffalo, while US 20 will take you around Buffalo.
From US 20, decide: Niagara Falls or Lake Ontario?
If Niagara Falls and the Niagara River is your choice, take US 62 N to NY 104 E, but turn off 104 onto NY 18 to get to Old Fort Niagara and the shore of Ontario.
If you want a straight shot to the Lake, from US 20 in Lancaster pick up NY 78 N, it'll take you to NY 18 at Olcott.
Follow 18 E to Lake Ontario State Parkway at Rock Ledge Beach, follow LOSP to I-390 in Rochester. Follow I-390 S to NY 31 E. Just after you cross the Genesee River in downtown Rochester, take your 1st right turn, go a block and stop for food at the Dinosaur BBQ (bikes, blues, booze & BBQ). There's always free up-front bike parking (and no, I have no financial interest in the place!), and I stop there when I'm in that town (there's a Dinosaur in Syracuse, too). After eating, retrace your ride to I-390 N to get to NY 104E.
OR from I-390 pick up NY 104 east. Stay on 104 'til you get to NY 104B in Mexico, it'll end at NY 3 N. Take NY3 N to NY 180 just past Sackett's Harbor. From 180 take a left on 12E, it'll take you to NY 12 and follow NY 12 to it's end at NY 37. Take NY 37 E to Malone, where you'll pick up US 11 N. That will take you to US 9 S in Champlain.
Follow US 9 south to home!
