Nope... I also have a transponder to get into my community in the other bag and it always opens. When I have my wife on the back I need to get closer to the gate as her leg is blocking it. But it still opens.
You Guys are the greatest! Even the questions I thought of after I posted were answered. So my plan is to put a transponder in a leather tool bag on the front of the handlebars. This is great news gang! Thanks to all who responded! ... Ross

As long as all of your vehicle tags (bike, car, truck, whatever) are registered to the unit, you're good too go.
I'm not saying however, that if you go through a Sunpass booth 100 times without the transponder, that Sunpass folks will not send you a nasty letter.
