Be careful with starting fluid. That stuff is pretty harsh and a little can be too much.

If you have spark at the right time, with starting fluid, it should have done something. Are you sure about the spark?

Does it sound right as it normally does when cranking? Not faster or make unusual noises when cranking?

Check the air cleaner for obstructions and also the exhaust. A bird nest in either will prevent a start.

I'd check the compression before removing the timing cover. As mentioned, you need air, fuel and a spark at the correct time for it to run. And even if the timing if off some, it should still backfire through the intake or exhaust.

Start with the simple things first to eliminate them and go from there.

Rather difficult for me to follow since it looks like a long sentence.
Reads like you have a 1999 Road King.
Unknown miles, unknown ownership history.
Unknown changes from factory stock.
In 1999 Harley had a recall or service thing about the bank angle sensor bracket.
The broken bracket would allow tip over switch to trigger and shut motor off.

Note:
That bank angle senor (tip over sensor) will reset if bike key switch is turn OFF/ON 3 times.
Consider review of bank angle sensor bracket just to make sure the bike does not turn off during a turn or bump in the road.
