sometimes yes, sometime no.
a 6 can be a "close ratio" with a 1:1 final
or it can be an overdrive with a .86 ( or so) 6th gear, 5th is a 1:1
andrews has charts as does baker drive train
andrews has a doc:
28_RatiosMPG-pg28.pdf
which I find very handy for ratios/rpm/road speed
depending on what you ride, what you are doing, overdrives can be achieved by a smaller rear pulley ( and doing away with belt guard) 65 tooth sometimes retains same belt ( had that on my evo bagger), 61 tooth requires a softail belt ( i have this on my FXRp)
or a slightly larger front pulley for overdrive

the new harley stock 6 speed is a 1-1 6th (just like the 5th in the older models)the make the primary and final drive ratios taller, and basically add a lower first, not low enough in my opinion.its worse on the 07-08 models the 09 up have a slightly lower final drive ratio,if your adding a six speed to a 5 speed bike i would recommend one that uses a true overdrive.

I've got a SE six-speed in my 06 Road King. The first five gears are exactly the same as stock, the sixth gear is an overdrive, completely extra.I use it only when I'm cruising at 70mph or better.Absolutely fantastic to be doing 75mph and turning the same rpm's as 65 in fifth gear.

5 speed - ’85-’93 (except ’93 Softail) - stock 32T front / 70T rear pulley - 3.37 final drive ratio ( Domestic models!)
5 speed - ’94-’06 (except ’95 Softail) - stock 32T front / 70T rear pulley - 3.15 final drive ratio
the .86 or 1:1 ratios we talked about are the ratios of the transmission only
final drive takes in the primary drive ratios + tranny + rear belt pulleys
