Transmission issues
Recently going down the road it took more effort to shift up...i don't remember what gear....plenty of clean trans fluid..while sitting still with it running going thru the gears I have all the normal harley clunk except from neutral to 2nd there is a grind along with the clunk. It's obviously different. If I'm in 1st and miss neutral it will go into 2nd normally with no problems. ... I guess the transmission needs pulled to replace synchros??? ..but I am really not a transmission guy and would like a better understanding of what is going on...my next move and about how much am I looking at.
Your trans doesn't have synchros, so don't pull it yet.
Two things you didn't mention- what is the year of your bike? There are differences in the shifter mechanisms.
Did you try performing a full clutch and cable adjustment?

Yeah I'm pretty dumb about transmissions. .. It's a 2003 wide glide. I didn't do anything to it yet..wasnt going to til I had an idea of what was happening. Didn't readjust clutch because I didn't think it needed it and also since it was just effecting one gear and not all equally.----im not saying I won't just why I didn't. I would love nothing more than to adjust the clutch and fix it.
