Yes, I usually apply ride-on sealant when I am ready to ride more than 5 miles, that's exactly what I did.
Have your tires mounted on the rims and wheels installed, then squirt the goo in recommended dosages from the Ride On website. You need to be able to put at least five miles on the bike within 30 minutes of applying the goo, so install it AFTER the wheels are installed on the bike. The initial ride distributes the goo as needed within the tire. You need to ride it soon after installing to prevent it from setting up.
