No, you don't have to take the weight off the tires. When I adjust mine, I never leave them fully turned to firm or soft.
I always back them off just a bit. It is kind of like a cheap faucet. If you leave the shock body jammed in one direction or the other it is hard to break it loose the next time. I also just barely snug the retention nut when I am done, because it tends to tighten more as you ride.
The shocks come from HD set at the softest setting, with approximately 4 threads showing. To firm them up you loosen the shock body. This extends the shock making the ride firmer. So it is just the opposite direction then you would logically think you should turn it to firm them up.
Have you been trying to turn them the right direction?
Once you get the retention nuts and the shock bodies to break loose and turn, from then on they are easy to adjust each time.
Tom

Taking the weight off often times makes it easier. If the bike has a lot of miles on it, or even if the thing's in a tight place, sometimes using a punch and hammer makes it a lot easier. You wanna use a blunt face so you don't chew up the teeth on the preload collar, but other than that, it can take an hour job and turn it into a 10-minute one.

run the lock nut all the way out and snug it up against the frame then use your spanner to adjust

if you look at that picture i cant turn the locknut up against the frame because of the head of the bolt stops it.

the outside of the shock body can turn, but the bolt should not. I'm sure you have soaked it with WD40 or the like right.
