i tried to adjust the preload on 2000 softail deuce loosen the locking nut but could not turn the adjusting wheel with the spanner wrench do you have to jack it up to get the weight off of it or what do you do.
No, you don't have to take the weight off the tires. When I adjust mine, I never leave them fully turned to firm or soft.
I always back them off just a bit. It is kind of like a cheap faucet. If you leave the shock body jammed in one direction or the other it is hard to break it loose the next time. I also just barely snug the retention nut when I am done, because it tends to tighten more as you ride.
The shocks come from HD set at the softest setting, with approximately 4 threads showing. To firm them up you loosen the shock body. This extends the shock making the ride firmer. So it is just the opposite direction then you would logically think you should turn it to firm them up.
Have you been trying to turn them the right direction?
Once you get the retention nuts and the shock bodies to break loose and turn, from then on they are easy to adjust each time.

Taking the weight off often times makes it easier. If the bike has a lot of miles on it, or even if the thing's in a tight place, sometimes using a punch and hammer makes it a lot easier. You wanna use a blunt face so you don't chew up the teeth on the preload collar, but other than that, it can take an hour job and turn it into a 10-minute one.

there is four threads showing when i try to turn it the whole body of the shock and bolt is turning is turning so i hold the bolt on the end of the shock then i cant turn it when the whole body is turning it doesnt look like it is moving up on the threads i am turning counterclockwise. Is the whole body and bolt suppose to turn.
