Is there power to the light socket? If not, check the wiring under the fender. Could have come loose from the bracket and rubbed on the tire.
Don't know what bike you have but mine has a TSSM (turn signal security module), not a flasher. Bikes without security have a TSM. Could have dirty connections where it plugs into the electrical caddy.

I just jacked the bike up a little to create a gap to work between the fender and the tire, spliced the bad wire, taped it, and positioned it away from the tire making sure it was in the clips to avoid future rub. That was last Summer and no problems since.

Before you tear into it, switch the bulb from the good rear to the bad ...that will tell you if its the bulb. Then before you start jacking the bike up, while you have the lens off, push on the silver insert around the bulb fixture, inside the turn signal housing. I just had the same thing happen. It's sorta like the wires are bad inside of the housing. Right rear of Street bob.
