needle clip position?
Since the Mikuni is not a "stock" carburetor for a Twin Cam, there is no "stock" needle position. You are going to have to start somewhere, read plugs an adjust from there. Final position of the needle will depend on what you particular build needs.
a mikuni hsr 42 comes with a size 25 standard pilot jet ; 160 size main jet ;and a j8-8ddyo1-97 jet needle. the jet needle has 5 grooves in it, with an e- clip attached to one of the grooves. just not sure which groove it was in, when it was new in the box.
Always the middle clip, what is your altitude? and engine mods.
If you have the tuning booklet, turn to the back cover where the parts explosion is.
look up part #61 this part is not commonly "tweeked" in this carb, but on a harley it should be switched to a slightly narrower one. This eliminates the off idle "sag" or dip that you commonly see on dyno charts with these carbs, and run the standard needle and you will probably be switching out the pilot jet for a smaller one. like a 20 or 22.5
