I use Navigon. I've had it for years. I started out not liking it, but they've really made it great since Garmin bought it. It's pretty pricey, and can take up space on your phone (preloaded maps), but I've used others that only work when your cellular service is good, and I've had that bug out on me too many times. There are also in-app purchases that I just bought yesterday. It really is a great app.
IF YOU GO WITH NAVIGON, THE APP (AND IN-APP OPTIONS) ARE ALL ON SALE FOR A WEEK OR SO, SO NOW'S THE TIME TO GET IT!!!
Waze is a good, free app... gives turn by turn, and community fed traffic/weather. It's what most tech reviewers like most.
Apple is coming up with their own (dropping google maps) that will have turn-by-turn this Fall, with iOS6. So you could use a free one until then and see how Apple's turns out...
I use Navigon on the iPhone with the XM Skydock on the EG & pickup. Works very good, dims music for turn by turn directions and you can install as many states & provinces as you want to keep disk space available.
Gee I wonder how the rest of us without the toys ever managed to go round the block much less the millions of miles that have been ridden without them ..................

Only reason I have a cell is job search related and my garage shop other wise I use the house phone to beep so I can find the thing some days ............ . Never had the battery die or needed to reset my Rand McNally .
Funny thing we all raise 6 kinda hell over cagers on cell phones but how many in here are screwing with a device while riding . Pot calling kettle .............
I use MotionX GPS Drive, the free version, you can purchase the turn by turn voice guidance for 30 days at a time for like 2.99 or something like that, witch is fine by me, I may only use it one week out of the year.
I travel a lot and use this ap; it really works well. I paid 9.99 for a year of voice. Works as good or better than my garmin did without paying for the unit or map upgrades.
I bought a RAM mount to hold it on my bike.

I use tomtom app in iPhone 4 in a Ram mount and a waterproof case. I can plug into bike stereo or Bluetooth to my helmet for phone calls, tbt and music (I don't use the phone facility). Silences tunes for instructions and then kicks back in.
I use both UK and US apps, prefer it to Garmin due to interface and mapping clarity. Updates included.
Point to note in general: make sure whatever app you choose doesn't rely on data connection to work - many do. Useless when out of signal. You need one where the mapping is stored on the phone.
I use the Mapquest app. It was free, voice guided, turn by turn navigation. It will interrupt your iPod use. I have a harming in the car and this is easily as good
