2001 ultra classic fuel problem

Check the connection going to the tank. Not sure if you have one or two fuel lines on the tank, but if you have the quick connect fitting the ball bearings in the fitting will cut into the collar, causing the nipple not to connect with the check valve. Take the connection off, and pull back the cover. You can see if the bearings cut into the collar. Try reseating the fitting, that will sometimes work.
How many miles are on it? Have you ever replaced the fuel filter?
If that doesn't work, then inspect the internal hoses. You can remove the gas cap, and turn the key on. You can hear gurgling if the internal lines have holes in them.
