On my Trip Planner, I got this...
I-10 to San Antonio
Then 90 in San Antonio
Then 385 in Marathon
9.5 hours and 600 miles.
Or
I-10 to Ft Stockton
Then 385
It's 9.5 hours and 650 miles
I've never traveled these highways/roads to I can't say how they are.

Have ridden both routes in Chick's post and option 1 is the better, IMO. I actually enjoy that fairly lonely stretch of road west of Uvalde. Maybe use the I-10 option on your return to come back through Fredericksburg and Bandera. Can't recall that exit off of 10 that is west of Fredericksburg but it wouldn't be a bad way back to Houston.
