Yes, but has no one told you that there is a Helium shortage? Might have a hard time filling your tires from time to time.
Using Helium is just plain silly, the molecule is smaller than air. It would just make the leak worse, which would help him find it faster. Or be stranded quicker.
No tyres, with or without tubes, are truly air tight - or nitrogen tight for that matter. The gas inside will slowly leach out. Which makes using Nitrogen seem futile to me. Just top up with fresh air!
OP's tyres may have gone down due to a leaking valve, or a slow puncture, who knows? Nothing is permanent where tyres are concerned, by definition!
Instead of Nitrogen, try Nitrous. I use it and it gives me an extra 50hp.
As for the leak, try putting a little dish soap in a spray bottle full of water and spray it on your tire, around the valve stem and the spokes and see if you get any bubbles. If you wanna get crazy, pull your wheels off and submerge them in your neighbors pool and look for bubbles.

it's 20.1 on our digital readout in the hallway at work. When in the room that we refill our nitrogen dewers it sometimes dips low to set the alarm off lol. 19.5 and your still good to go. Hit 19 breathing is getting pretty tough.
Dont want to sound stupid, so correct me if I'm wrong, but your using nitrogen in a spoked wheel !
Have you sealed all the spokes then or what ?
I thought the big argument about using tubeless tires with spoked wheels was that invariably you always had a big chance of the tire walls not seating good and leaks around the spokes !

that is why I said "approx." ...our confined space meters go into alarm if the o2 levels fall below 19%....and depending on what I had for lunch the methane levels may be much higher than is normal and healthy for a confined space...
