I just bought a new 2021 Road King. Put on Vance and Hines, naked breather that fits under sock air intake, Vance and Hines Dresser Duels, and Vance and Hines Tourqer 450s Destroyers. Very confused by exhaust sound. No loud idle rumble. If I get on it, 3000 plus rpm it sounds great, but otherwise it sounds like a scooter. At low RPM the decel is loud and rumbles, but acceleration sounds like scooter. I did not yet retune. Could this be the issue? I dont want to take to Harley to retune but the Fuel Pack 3 wont have 2021 maps until December and the Dyna Jet does not yet have maps either. Would pulling the packing out of the Vance and Hines Tourquers make a difference?

Yes, it probably would reduce your low end torque, but at least it would be a bit louder.
My opinion is that the 450s arent going to be that loud, stage 1 or not. To me the 450s have a cloudy sound in the lower registers. 400s would likely be a bit raspier, but you'd have to add decent cam to experience a significant increase in sound.
And - not having anything to do with sound - the bike needs a proper tune for stage 1 mods.
At this point we know you aren't concerned about the warranty so I would either contact Fuel Moto or Thundermax I know they were both almost ready to release their tuners for the 2021 bikes

Your description of the 450s sounding cloudy is spot on. Ill pull out the packing and see the difference. I dont want very loud, but would certainly like a deep rumble. I understand it is now running lean until I can retune. Is excessive heat the only issue to be worried about? Im worried about riding it until the tuners become available for 2021s. Im running at 290 degrees shot with a hand held temp gun right at the bottom of the exposed plug on back cylinder.
I would get the FP3 with the stock map and let it auto tune. That will at least get it running properly with the stage 1 and not cause any future damage as you are probably running too lean.
If the sound does not improve to your liking, then a decat of the header pipe may be in order or look at a new V&H header pipe as a replacement.
