I'd have to say that it's a tie between Buford and Will2002. Since I live in CO and ride the great Rocky Mountains I am definitely partial, but dayum...the mountains are incredible from Glacier National Park to Southern Colorado.
However, the ride from anywhere to Sturgis is definitely one you'll never forget. On top of the ride, you will meet people along the way and during Sturgis that will become lifelong friends!
I have a group of people that I in 2005 (through 2008) that have become a second family for me. We all stay at the same place and generally ride together the whole week, day in and day out, go back to the motel and then drink and shoot the $h!t til the cows come home.
I want to put the miles on this year and I need to know where the best ride in the USA is
Alot of people have said that Americade is the best bike run of the year. It's the largest in the Country & happens in Lake George, NY June 1-6. May give it a try one of these years?
The Rockies especially from Ouray, Colorado thru Silverton, to Durango over Red Mountain Pass. U.S. 550. Also many parts of the Pacific Coast Highway are my favorites. Ain't it great to ride in the U.S.A.?

Best short run, for me, has to be Needles Highway in the Black Hills. Best run of any length would be Dover, De to Pittsburgh, Pa by way of 896 to Lancaster then onto 22/422 across Pa.
Best short run for me is up Route 7 from Brookfield to Geneva. Just when the sun is starting to set on Lake Erie. Its relaxing no cell phones or problems just me and CCR on the radio and the wife.
Don't let them kid you the smokeys are not mountains the mountains are in Colorado. Independence pass, million dollar highway, Estes park just to name a few that are over 12000 feet. Independence pass is over 14000 feet high, above tree line with glaciers. Colorado has some of the highest highways in the states and some of the most beautiful. Once you have traveled the mountains of the west you will laugh at the little smokeys being called mountains. There are some beautiful highways in the Smokey hills it is worth the trip to see but don't expect real mountains there will not be any snow on them in July. There are some beautiful roads in the east. But in my opinion you need to see the Rocky Mountains it will be a ride you will never forget. There is nothing like it. JMHO
This past summer I rode from VA out through South Dakota and the Badlands to Mt Rushmore, Deadwood, Sturgis, then up into Montana and over to the north entrance of Yellowstone and all the way through the southern end and into Jackson Hole, WY then up and over the Grand Tetons into ID, UT, CO and then back home. If you ever get a chance to do a ride like that you will see some beautiful country.
Hey, I'm in your neck of the woods (Northern VA). How many days did it take you on this trip? I'm planning on going west next year and yours is very close to my plan.
Trail Ridge Road in Rocky Mountain National Park in the hottest part of summer. Yellowstone's OK, but the traffic is sometimes really terrible and if you hit a construction zone when it's raining, you're screwed!

From about Redding,Ca to Canada the mountain highways east and west from Nv. west are some of the of the most beautiful mountains in our country. They offer some great riding roads and the most incredible views. You can start on the east side of the Sierras and just keep going west to the Pacific ocean go north a little thee go east over the mountains go north a little and repeat until you are at the Canadian Border.
