Import (upload) the route (GPX track) to the H-D Ride Planner.
Zoom in and inspect each waypoint to make sure it's on a road segment, not to the side in some building or on a beach, not in an intersection, and if on a divided highway on the right side (direction).
Make sure there is a start and end destination (forget off-hand what they call it).
Save it to the Ride Planner cloud.
Load up the H-D app on your phone, scroll to the ride and make sure it's there.
With the phone on the USB tether or connected by bluetooth, locate it on the GTS head unit.
Ride it.
Google maps is fine too, but has its own problems; like if you create a route on a laptop or tablet any route drags are lost when it's sent to your phone. Conversely Google Maps on iPhone doesn't allow route dragging. Traffic information and exit information both require an active connection and a reasonably fast one at that. Can you even load a GPX track on Google Maps? I've never tried and don't know how well that will work.
And I might add...make sure to place your waypoint just after the intersection on the road you want to turn onto. This way it forces you to make that turn.

This! I use Tyre with this method and rarely have issues. When I do, its because I failed to double check the route in HD Ride Planner.
To those who say use Google....its primarily a Point A to Point B GPS getting you there the fastest way possible. I use it when I am not on a trip where I am trying to go an explicit route.
Agreed. Google Maps is great for getting from A to B...not so much for routes. I'm sure there are others much smarter than me but I have not been able to figure out a way to create a route (the way I want go) on my desktop and then have it transfer to my iPhone to use in CarPlay on the bike.
So I deleted my Ride Planner app because it wasn't working and I see it's not available as a stand alone app anymore, it's integrated into the Harley Davidson app. How do you get it (ride planner) to show up on the GTS? via carplay? I have the Soundstream HDHU14+ and I'm not seeing the Harley app as an option on carplay on my phone.

The H-D app isn't supported under CarPlay. Apps have to opt-in and be accepted for CarPlay use.
