How about if I'm going to turn the engine over with the ignition off by rotating the rear wheel. Should the wires be grounded? My instincts say no but they've been wrong more than once.

if you have the ignition on you should ground the plug wires on the bikes with electronic ignitions. don't worry about it if the power is off. you could also pull the coil wire off to kill power to the coil.
