I-57 from Chicago to Sikeston, MO and then I-55 south would be your fastest way. It's been a couple of months since I was on I-57 so I don't know the construction update for I-57. I was last on I-55 about 2 weeks ago. I-55 has had ongoing construction from about the Troy exit with traffic sometimes at a stop for several miles. Also some construction on I-255 near I-64 junction.

US 51 is an OK route the entire way but I can't say on construction. It is quite a bit slower tho going thru towns.
Thanks for the suggestions guys....was going to leave at 8am today, but some bad storms in southern IL have pushed us back a few hours. Hope to get out of here by lunchtime.

If you haven't left yet or are checking things along the way, I've always enjoyed the Great IL River Road back from StL, Alton, 3/100 along the river and you can ride along either IL or Miss rivers in some parts.
Be careful along 3 at night from Chester to Cairo...I came back from there a week ago and the floodplains are all lakes now, which means it will rain insects in merciless fashion for 2+ hours!!
If you can do some Ozarks on the way back it's fantastic from Ark to MO and thru the Twain forest.
And finally, I just found a kickass iPhone app called GreatestRoad just for bikers. You can rate, review, follow others' routes and pinch in or out of the maps...
Cool. I'll look into that route and app. We are in marion,IL now and a huge thunderstorm is heading toward memphis, so we are going to hit the local Harley dealer and may go to bowling green,ky for the corvette museum.

You could also come down the peninsula between the rivers
into Brussels Ferry and take the free ferry across the river and south
into Alton.
Keep in mind Hwy 3 is pure crap from E. StL for at least 10 miles. But after
Chester it gets
great again. There's a stretch of bluffs & hills that is stunning just east of 3,
search for Bald **** Cross. Pretty country in them hills. 3 is a floodplain, but just east is more interesting.
Cairo is plain creepy. Army Corps levees/gates on both sides of rivers, and a pure GHOST town in the old historic district. Ride around the cobblestones for a couple minutes past the old abandoned theatre etc...
