Where I come from Any Knock coming from a motor is No Good and usually means Problems. Since the bike is a 2000 it's probably out of warrenty so I think you should take it to a dealer / independent, Have them diagnose it and then go from there. Not much help I know buts thats how I would probably handle it.

most engines will knock a little when cold. the severity of the knock depends on mileage, engine wear and oil vescosity. not really much to worry about, but you may want to let the bike warm up a little before you gag the throtle!!

Had a similar knock on my Softail. Turned out to be a loose nut on the compensator sprocket in the front of the primary. You have to pull the outer primary cover to check it and when you pull the outer, you need to replace the gasket as they are a one shot deal. Good luck.
