What year and model of bike? Carbed or injected?
start with the simple things first, plugs, wires, fuel filter etc.

That may be the problem ... it should have 2 cams. Sorry, couldn't resist 
Have you confirmed spark at both cylinders?
Conpression test?
Bad injector?

The bike runs like STINK! Fastest Harley I ever sat on!
Do your bikes "load up" and even backfire through the exhaust when you give it gas from low revs and the motor is hot? I have to keep my revs up, then everything is OK. Starting from a stop I have to give it more revs then use the clutch gently...
Is this happening on your bikes? Seems a natural, given a cam that moves the power band up.

Sounds to me like the tune is lacking.... needs some work...
