I have had 2 sets of sealed spoke rims. I had 18" 80 spokes on my '99 RKC and the wheels on my RG are also sealed. Over 10,000 miles on the RKC and never had a problem. In fact it's still running around and it's on a second set of tires. Guy I sold it to said he didn't need to reseal.
Only have around 5,000 on the RG, but no problems yet. I imagine if a spoke did come loose, the wheels would have to be re-sealed. If it happened on the road, I would just put a tube in to get me home.

I saw ina mag the other day about a company that makes a billet wheel, that gives the appearance of spoke wheels. I can't remember the name of the company, but it seems like that would elevate some of the issues people have with spoke rims.

i had a setof 60 spokeon my 98 flhtc, i think they when from american wire & wheel but cant remember for sure. anyway, i wasrunning down the road about 70mph and the bike startedfeeling weird. i let off the throttle anddidnt feel anything. throttled back up a little and the rear of the bike started going up n down big time. i let off throttle and eased the front brake on slightly to pull off the road. looked at rear wheel and 1/3 off my spokes went through the seal into the tire. pretty scarey ride!!!the were hardly over a year old when it happened.
