Good suggestions so far, I have been working on this all day and I pretty much have the trip down figured out. We are leaving home(KY) and riding down through TN on HWY 111 to I-75 at Chattanooga and we're gonna run 75 down to Macon GA the 1st day, it's 350 miles for us. Day 2 we leave Macon GA on I-16 to exit 51 HWY 441 and follow it all the way to I-10 at exit 303 and run I-10 into Jacksonville for night 2, it's 275 miles. Day 3 we leave Jacksonville on HWY 13 down to St Augustine to see the castle and the town and leave St Augustine on HWY 207 down to HWY 17 and follow it to Deland and cut across HWY 44 to New Smyrna Beach and down as close to the Space Center as possible. Day 4 we are going to check out the space center then head south to Melbourne and cut across 192 to get to 441 and follow it down the east side of Okeechobee then hit 27 and go on down to the Everglades, don't know how far we'll actually get this day, depends on how long we're at the space center. Day 5 or 6 whichever it ends up we're going to check out the everglades and if everything is running smooth at that point we'll decide on going to Key West. I have looked hard at HWY 27 to run back up through the state, seems like a decent choice. If time allows when we get back up to northern FL we're going to run around the gulf coast to Panama city and back up through AL to home. We have set aside 2 weeks to do this trip to allow for weather and problems on the road, I don't know if we can stay away from the kids that long though. Keep the suggestions coming, my route isn't a must, I'll gladly change it for a better trip.
A1A (east of Jacksonville) thru Ponte Vedre Beach, over the Vilano Beach bridge (look south when you're at the peak, looks like you're riding into Spain). Stop and tour the shops. Go east over the Bridge of Lions back onto A1A south all the way to Flagler beach. At Hwy 100 look for turn to Old Dixie Highway. Great little scenic ride. Puts you out into Ormond Beach just north of Daytona. At Grenada Blvd, left over bridge back to A1A.
If you come down the west coast of Fl, take US hwy 19 to Hwy 24 west of Gainesville. Ride out to Cedar Key. Great food (especially seafood and signature crab bisque) at Froggs Landing.
Any way you go, have fun and be safe. I'm looking forward to Spring to ride from Jacksonville back up to the Smoky Mts

Sounds like you have the trip down decided but I would recommend 41 for the ride back up the west coast. It gets a little crowed between Ft. Myers and Bradenton but is a real nice ride from Bradenton to Tampa. North of tampa you can take Hwy 52 over to 19 which is a nice 4 lane with just a few towns to go through all the way to Tallahassee and get on 27 to Bainbridge, Ga, Left on 84 to Dothan, Al and pick up 231 to Montgomery where you can get on I-65 if you have had enough or just keep going up 231. This is all great riding and if you get around Sarasota give us a Holler and We will buy Ya'll dinner. Happy Trails and ride safe.
I envy you. We now live in Atlanta and wish I could ride the Florida roads once again as we are from Jacksonville. If you get the time in St. Augustine, look up Poorboy there. He is on this site and rides a FB. Great guy and a lot of fun to be around. I am sure he can provide you with more routes to and from the Keys.

Oh yea, before I forget, Florida is F L A T, not like Georgia and Tennessee. I do love that part about being up here; HATE the cold weather though. We spent Christmas in Florida with our Son and Grandson and the weather was 80 degrees every day!!

Thanks Just Dave, people like you are what makes this such a great site. We rode 19 and 41 on our last trip so we're probably going to go back up more in the middel of the state. Once we head back up we are coming around 41 timiami trail and check out some of the stuff around the everglades and then it just depends on how soon we need to get back home from there, we may have to run some slab to make up time but if not we're going to wiggle up the middle of the state.
