I ride solo and yes it is a street glide. Have a whopping 1300 miles on it so far. I checked all the bolts last week on the rear fender and the fascia and all seemed tight. Have not seen any rubber coming out from anywhere.
I am determined to find the issue though. All great responses and ideas. I will try the exhaust bracket first and then dig deeper into the fender area. The rear signal and license plate light seems tight as well. Grrr. Thanks again.
remove your saddle bags and check the supports and rails. Supports sometimes break at the bottom where the nut attaches to the rails and the rails get loose at front mounting area.
On mine, I found that the trim piece around the rear fender that the lower light is in was vibrating against the fender. Had to loosen and reposition the whole fill panel to get rid of the rattle.

I have the same noise on my FLTRX......At first, I thought it was something internal on the mufflers, because the sound seemed to match the frequency of the exhaust..... but now I think it's the plastic fascia, or maybe the tri-light. Went over every nut and bolt back there, and it's still there.
Well at least I am not the only one to have this issue. Tonight I am going to take the bags back off and check things out again. Everything seemed tight but may it isnt. Just keeps getting worse to.
Also check the Jubilee clips that hold the chrome exhaust covers on, i had one that had snapped and the cover was vibrating against the pipe. Just a thought?

I used painters tape, and started taping seams and stuff...... conclusion: On mine it's a combination of the left side of the fascia, the license plate frame, and the protective cover over the tri-light.
I'm thinking clear silicone sealer from the inside of the fender, and some foam rubber under the license frame.
