Ride-on use
I personally wouldn't take the chance. If you want to run tubeless on spoked wheels, there's a company called Outex that makes a kit using heavy-duty rim tape that is supposed to seal the spoke holes. Here's a link. No idea whether they are reliable etc.

Ride-on will only seal punctures in a narrow portion of the tread area.
Not all spoke rims can be sealed. Even if the spoke and valve holes can be sealed, the bead shoulder must be very smooth so the tire can seal on the metal. For safety, the bead shoulder also needs to have the raised bead retention band on the inboard side of the tire bead to prevent the tire rolling in on itself if air pressures drop.
