As others have said, relax & don't grip so tight...your bike will also handle better as a result.
Also, the new FXDL has smaller grips than all other Full sized HDs....this could be compounding the problem as well. Swap out those skinny grips to a set of regular diameter grips, like are on all the other Dynas. This was a bonehead move on HD's part IMO.

The new FXDL I bought had terrible little grips on it, those were changed out day one.
iso grips, throttle boss ( on both sides). figuring out how to use the throttle resist doohicky to work as cruise control to rest throttle hand. clutchhand in lap every now and again.
I use a throttle assist that is part of my Avon grip, makes a ton of difference. I often find myself riding with my hands completely open, fingers over the lever and thumb just hanging out, using my palm to do all the throttle pressure.
Does anyone have problems with their thumbs? I have had shots operations and acupuncture ,now just take pain drugs.Looking for an orthopedic gloves .This hand problem has stopped my competition shooting and don't want to stop my bike riding. Drs. say you must live with it.Getting old sucks. Hard work has really paid ,body needs replacing. Any help would be good. Going to try riding with shooting gloves next long trip.
Crampbuster from Amazon. The large one in chrome was less than $20 shipped and is a TERRIFIC device that eliminated cramping for my right hand. Cheaper than new grips and solved the issue for now. HIGHLY recommend!
