You will know if a spoke is loose if you tap it with the wrench a it sounds like a dull thud. When working around the wheel, tap each spoke, and if you have the spokes torqued properly, you will hear a consistent ring tone from each spoke when you tap it with the wrench.

I feel stupid, what I thought were loose spoke nipples, are actually just my wheel weights from balancing, that was what i saw. Still a good idea to check my spokes though lol

No. think of the spoke as a long, thin bolt, and the nipple as a nut. The spokes are standard right hand thread, so you turn the nipple clockwise to tighten, like any nut.
For Harley-Davidson spokes, youll need an 8 gauge spoke wrench. I bought mine about 40 years ago so I dont remember where it came from.
