I have a 2019 street glide. I put on a S & S air cleaner, Rhineheart 4.5 slip on mufflers and a Fuelpak tuner. Are these modifications compatible to use for a stage 2 kit?

You may find limited support for some cams with the Fuelpak for your tuner. So when selecting a cam, you might contact the vendor or Vance and Hines about a map for the cam you choose before purchasing it. And the Rhineheart 4.5 may reduce the torque available at low RPM's. If that is the case, you may benefit from different baffles.
Even if your 2019 is low mileage, I'd recommend replacing the lifters, cam bearing anjd flywheel runout. And I'd check to see if the latest oil pump is installed in your engine while things apart. The latest HD oil pump has an additional oil seal that helps prevent sumpping. I believe they came in the later 2019 models, so depending on the build date it may or may not have the latest pump. The HD pumps are fairly inexpensive and good quality.

I would seek a better tuner and have it tuned on a dyno by a competent tuner. TTS Mastertune or PV. Whichever your tuner prefers. Most professionals prefer the TTS. I don't know too many tuners will tune with the FP.
Also, the latest HD pump, while a step in the right direction and rekatively inexpensive, has still sumped in some BB builds and there are better, tho pricier options. It's probably fine for a stage 2 depending on your riding style.
If a full on BB build is in your future and you like to hotrod, I would consider a better 3 stage pump and plate. JMO.
I don't know about the FP4 but the FP3 can tune an S&S 475c cam. After starting with their canned tune, run autotune, then send that tune to V&H and have them refine it. I ran the 475c on my Fatboy 114ci with Big Radius and after the last tune she ran great. I never had it dyno'd on a machine, only by ***-dyno. Don't let anyone tell you that you can't do something or that you'll lose this or that, unless they themselves have tried it.

How would you know what kind of tune you have? Your butt dyno? You have no clue what's left on the table.
I forgot that you seem to know what's best. If it's good for me what does it matter to you if something is left on the table?
Thats fine. I could care less. But you can't say it works til you can say it works. I call bs. Prove me wrong. Until you can...you cant.
There's an FP3 sitting in the circular file here if you want it. The 98 build sitting here isn't going out without a good tuner. It's not just power that gets left on the table.
All BS aside . Cam and exhaust must work together . Wide open exhaust will not play well with a strong torque cam.
The VH tuner is limited it what it can do and No tune we fix the wrong exhaust with the cam chosen.
You have some home work to do and the answer is not in a format like this. This is where you find the question you need to answer.
Go to Fuel Moto sight. Look up what you have and how does it work with the different cams.
DYNO tune is the least of your concerns at this point.
Again no tune will fix a mismatched Cam and exhaust. You must know what it is you are looking for. Do you want low end strong torque. Do you want wide open power on the top . Maybe strong power in the mid-ranges you operate in. Until you know that. keep looking. Good luck with you search.
