I hope you make a run on Rt. 97 out of Port. The Hawks Nest is very well known. I'm sorry to read about the Kinzua Bridge, That was an awesome stop and walk. The worst motel I ever stayed at was on Rt. 6 in Towanda. Bear's H-D in Honesdale always had the best prices. (I'm a NJ boy)
Now look at your map. At Rt. 6 in Hawley is 590 heading east toward Kimbles which requires a turn south. This road follows the Lackawaxen River to the Delaware. At the confluence of the Delaware and Lackawaxen rivers is a Zane Gray museum. He wrote some of his westerns right there. There is also a very small bridge known as the Roblings bridge that will take you across the Delaware.
Anyway that road along the Lackawaxen is one of the best roads in eastern PA. And 97 out of Port is a rip. It would be a great way to start or end a tour.
Are you taking 32 north to get there? Have you done the New Hope thing? 519 on the Jersey side? It's all good.
Hearts Content Scenic Area in the Allegheny National Forest is a stand of old growth forest. The road in is gravel and chip seal. Not good for your belt and the tar never comes off but maybe you like old tall trees.
If you've ever been to the real thing, the Grand Canyon of PA is highly over rated. But it is on the way so take a look see.

Zippo bought out Case knives a good amount of years ago. Zippo and Case knives are in the same building as far as a the museum goes. The manufacturing of both are within close proximity of each other. You may also want to check out the oil well museum heading into Bradford. There is at least one casino a few miles up 219 in New York if interested. There are several Indian Reservation around there to.
We left Wellsboro between 8 and 9 and went to the Grand Canyon. Great ride but had to wait an hour or so for the fog to clear. Beautiful when it is clear. Worth the wait.
Lots of good ideas already mentioned. The best riding on 6 is west of Mansfield & route 15. You can Google & map out:
-- PA Grand Canyon. There are 2 lookouts. One is right on 6. (The other one isn't so far out of the way that you shouldn't go there too.)
-- Kinzua Bridge. Go in from 6. When leaving, turn left & go AWAY from 6 & go to 59.
-- Kinzua Dam. From the bridge, it's on 59 west. After the dam, 59 runs into 6 again.
-- Burnin Barrel near west rim of Grand Canyon isn't a bad meal.
-- Ox Yoke in Galeton is a much better burger than Burnin' Barrel.
-- Ox Yoke has little cabins too. From Port Jervis, an easy day ride to see these things, and get to Galeton & Ox Yoke before dark.
-- The Harley dealer near the canyon is Larry's. About 5 miles west of Ox Yoke, on 6.

Do a Google search on this stuff, and then you can map it out. When you head back, you should ride route 120. Some things to see also, a couple of GREAT burgers on 120, and as nice a ride as 6. Maybe nicer. From St Marys to Lock Haven.
Geezr won't steer you wrong. There is a place in Coudersport that will allow you to park your bike under a canopy. Can't recall the name of the plae though. Stop in the little diners along the way. Quaint and nostalgic.
Once you're in Wellsboro, take a little side trip down PA 660 toward Leonard Harrison State Park. There's a nice (and free) overlook of the PA GC. If you're looking to camp, there's a state park CG right there (water, electric,) and there's also Canyon Country campgrounds just before LHSP (RV park with nicer amenities.) Rt. 15 between Williamsport and Mansfield is a pretty great ride--it's like an interstate with almost no traffic that just goes up and down hills and valleys, just some great scenery.
Rt. 414 is a really nice ride too south of Rt 6, plus another nice campground right on the Pine Creek: http://www.pettecotejunction.com
