There is a Harley 6-speed OD conversion for your bike, plus the suggestions above. Baker offer both OD and DD, versions, direct drive - my personal choice for my old 5-speed.
If you are talking about using a 2007-up 6 speed gear set on your 2001 road king then no it will not fit. You would have to buy the screaming eagle 6 speed gear set designed to fit in your 5 speed gear box. Mixed reviews on it. Baker has better reviews for the gear sets but depending on how much you want to spend.
Another option is change out trans sprocket for more teeth or rear wheel pulley for less teeth. That will drop rpm. All depends on your end goal.
Got it. The research I've done, (internet), is that the Ultima is the kit to use outside of Baker, Jims, PRIMO, and Harley. I have seen a few OEM 6 gear sets for sale on Ebay and thought I could go this route. I have 6 speeds in my other bikes, and love the extra gear when burning up miles, and would like it in the Road King I'm having rebuilt to 98".
My 01 is a 98" and while I thought of going with a 6 speed, I can't justify the cost so that I can drop 500 RPM's at highway speed. But I don't have bottomless pockets or I probably would go this route.

Even easier and cheaper is a 27 tooth compensator sprocket
nets an 8.4 percent reduction in rpm
hesitant to bring it up as the op asked for 6 speed advice and seems willing / able to pay for that option

I would suggest there is a reason the Ultima kit is 5th in line of those kits.....
If it were my bike, I would go with a Baker gear set kit, or leave it as is....
IMHO..... the Baker kit is much better than the others on your list...
