Not assuming anything but what do you think the PV is doing to only affect one cylinder? A device that basically sits dormant until called into action to flash a map.

It’s just the Power Vision. I read only the piggy back units were prone to issues. I never saw were the flash tune units had problems. Bike has headwork cams throttle body and added compression. I don’t want to chance riding it with out the custom tune.
Just spitballing here…
Check the plug wires where they touch the gas tank. After learning that on HDF, I checked mine and the rear plug wire was almost worn through.
That’ll cause intermittent spark by shorting against the tank.

If you don't have the Target Tune module, the PowerVision is just a flash tuner that can also be left attached for a gauge... If you have your PV attached and using it as a gauge, remove it until you figure out the issue.

The worn spark plug wire from rubbing on the tank is a known issue, and a good call....... but I believe the OP already replaced plugs and wires...
I put new plugs and new wires on it already. That’s the first thing I tried with no success. I’m thinking it’s electrical as said, because fuel related probably wouldn’t effect just 1 cylinder as said. I had the pin hole in fuel line on my fat boy before, but you could hear the hissing from inside the tank with cap off. I ruled that out no hissing or bubbles in tank. I will take tank off and check coil and rear injector and connections as mentioned.
