Unless you have some severe wraparound type fender you may not have to mess with the wheel at all.
The four bolts that hold the fender on are sorta surrounded by these metal locking pieces. I can get at the front bolts, but the rear ones have proved to be a bitch.

Reach back there with a long slender flat screwdriver and bend back the tabs. Might be easier to turn the wheel hard in either direction and reach in from the backside.

It can be done. Good luck!

Still what kind of bike and did you buy the bike used or new? do you have a camera?

It's the 2002 Road King Classic in the background of the picture of me holding the end cap I got off after finding the set screws.

When I took mine off my 2001 RK one of the caps had the Allen head rounded out, was able to drive in a larger size and get it out. I then took one to a local nut and bolt shop and got some new ones that are a little longer, they stick out of the cap a little but give me enough to get a pair of vice grips on them if they give me problems in the future.

Mine were loose and rounded out by the techs that replaced My tires. I got the set screws out and replaced them with Machine screws. Easy on and off. Using the right length screw, You won't be able to see the head of the screw.
