Should a standard 5 speed mainshaft have any rotational 'play'?

Default gear lash
My '91 Flhs had gear lash same as my '07 Road King. Especially noticeable if you are coasting with the clutch lever in, then let it out quickly. Gives an audible clack.

Look on the side of the gas tank...if it says " Harley Davidson" anywhere the answer is yes! All kidding aside I have never seen a HD without some driveline lash. Mine has very little but still some and I'm sure as it ages it will have more.
