Knocking sound coming from rear wheel

Ur tranny to rear wheel isn't 1-1 so IMO that would rule tranny out.
Thinking I'd start with as above said,, jack her up and spin wheel forwards and backwards to see if its the same.
If so then U should check Ur brake system and rotor, make sure they are all tight and pads not worn out.
Then if Ur brakes are ok I then check to make sure nothings rubbing back there.
Then at last I'd pull Ur wheel and have bearing checked out..
Also check Ur belt and rear pulley to make sure nothing is embedded in either one of them...

Rear wheel noise
Hey gang -
'05 Fatboy started making this weird knock with every 1/2 rotation of rear wheel. In my case, the sprocket side bearing was teeter-tottering in its seat and had mushroomed the spacer slightly....drove my local tech guy crazy looking for the source!
I'd swapped out my original solid wheels for chromed ones last year (new bearings installed then), had noticed a weird "ping!" from the rear sometimes when I'd put weight on it, just sitting there, but not all the time....was totally fine otherwise. Almost exactly a year later, we were riding 2 up about an hour at highway speed, exited for gas when we noticed the steady knock at low speed.
Anyway, hope this helps someone out. Cheers!
