Just picked up a 2001 Road King and the rear cylinder doesn't fire. Has good spark(new plug) and compression. Plug appears to be dry. Can there be fuel getting to the front cylinder but, not the rear cylinder? Only 7500 miles. Any ideas/help would be appreciated. thanks, gregjo1948

As mentioned, we need more info..... It's hard diagnosing an issue via the internet. We don't know your knowledge/experience level, what has been done, and how it was done...
First, how did you verify the spark...? An inline spark tester? Or are you assuming good spark just because of a new plug?
Yes you can get only fuel to one cylinder if it's EFI.... Is the bike carbureted, or the Magnetti Marelli EFI system...?
Common issue for that era, the wire harness going to the injectors was a bit tight, causing breakage inside the insulation. Easy repair, solder in a 1/2 wire extension @ the break. This eliminates the tight spot, and repairs the broken wire that caused the issue.
