I have a 2017 road glide and I am replacing the right side cover. Tappet cuff for front cylinder stripped. I will be replacing the cam/cam plate/pushrods (adjustable). I will be stripping motor down to nothing. My question is if I know the length of the one piece pushrods can I adjust the new rods and put them in while heads are off and not have to adjust them again as if they are the original rods. Intake is 10.301", exhaust is 10.531". I can't see why not but just want to make sure.

Yes you can.
I would use it as a starting point.
Run the bike for a while, then I would re-adjust them to get the exact amount of preload on your lifters.
Adjust on a cold engine of course.
