As Matt stated, you do feel a "thump" when the top o-ring seats.
Before that I put the front cylinder on Top Dead Center by watching the lifters: exhaust up/down then intake up/down then plastic straw in front spark plug hole keep rotating wheel same direction until straw extends to the maximum - TDC.
For S&S prs with the S&S lifters that have the HL2T travel Limiters you extend the prs until they are seated on the lifters and rocker arms then extend them 4 more turns. Basically the first 3 turns take the space left in the lifters ~ .085", then the next turn will start to open the valves(you can really feel the resistance go up when you start to open a valve). This is because my lifters are not "pumped up" with oil - I had dumped the oil when I dissembled my lifters earlier. If I had "pumped up" lifters, the 4 rotations would be opening the valves right away. I could just finish adjusting my lifters now but I thought I would wait a bit to be extra safe.
To adjust the prs I will loosen them until they are just moveable with finger pressure then loosen 1 full turn(6 flats) then tighten the locknut down.

Got my prs in and adjusted, must say I really like the design of the S&S prs - simple to adjust and work with. Those push rod clips were a bit of a pain but doable, for me the exhaust clips were much easier than the intake ones. I waited to install the rocker covers until after the prs because it was nice to see the prs hit the rocker cup to be sure they were in the right spot. Anyway, making progress but rest of week is sketchy for time, have a lot of family stuff coming up.
