When I had the o-ring I reused it many times. I never had to replace it because it was leaking, just because it was 4 or so years old. Then they gave me the big gasket. I've taken the derby off at least a dozen times and have not replaced the gasket. Just don't over-torque the derby bolts.

The whole point of having a lubricant in the primary chaincase is to lubricate the chain and cool the clutch. The level of the fluid must be above the chain where it engages the clutch sprocket in order to accomplish this. If you get too high a level, it may cause the clutch to slip. If you get way too high a level, the clutch may drag as it tries to work in all that oil. The number of ounces you put in does not matter as long as the correct level of fluid is achieved.
